Abstract:
      A comprehensive analytical method based on ultra fast liquid chromatography tandem triple quadrupole mass spectrometry(UFLC-QTRAP-MS/MS) was developed for the simultaneous determination of 17 ginsenosides in different stored samples of Ginseng Radixet Rhizoma Rubra.Under the optimized chromatographic conditions,good separation for seventeen target compounds were obtained on a SynergiTM Hydro-RP 100 A column(100 mm×2.0 mm,2.5 μm) using 0.1%aqueous formic acid-acetonitrile as mobile phase at a flow rate of 0.4 mL/min.The target compounds were analyzed under multiple reaction monitoring(MRM) mode with an ESI source operating in negative ion mode,hierarchical cluster analysis(HCA) and principal component analysis(PCA) were used for data processing.The results showed that there existed good linearities for the 17 components in the certain concentration ranges,with their correlation coefficients larger than 0.999 0.The precision,repeatability and stability of the method were all satisfactory.The spiked recoveries for the compounds were between 96.4%and 103%,with relative standard deviations less than 5%.The results showed that the optimum storage conditions for Ginseng Radixet Rhizoma Rubra were as follows:storing it with a carton at -20 ℃,protected from light.The proposed method is useful for assessment and control on the quality of Ginseng Radix et Rhizoma Rubra,and provides a scientific basis for choosing the most suitable storage condition of Ginseng Radix et Rhizoma Rubra.
